Why I made it

In the lab I need to add a tag to the end of a protein in human stem cells. Prime editing can do that without cutting both strands of the DNA, but designing the guides by hand for every target is slow and easy to get wrong.

How it works

The pipeline designs and ranks prime-editing guides using PRIDICT 2.0, and picks a mode from the size of the insert: standard PE for short inserts up to about 40 bp, tethered-junction prime editing for inserts up to roughly 500 bp, and Prime Assembly for anything larger. It writes a text and CSV report for each run and runs in batches on the Imperial HPC.

Where it stands

The repo is private for now, but the designer runs live on the web at pegrna-designer.vercel.app and is still in development. If you work on prime editing and want to talk about it, use the contact form. The game uses a toy score and is not PRIDICT.
